The term Manglik comes from the term Mangal or Mars. Manglik dosa is the affliction due to the earth Mars in connubial life of an entity. It said in the Book of Vedic Geomancy when Mars sits in 1st, 2nd, 4th, 7th, 8th and 12th house with respect to Lagna or Ascendant, Moon or Venus the person becomes a Manglik. If a Manglik marries a non Manglik either there can be death to the other person or separation in that relationship. 
 
 From my experience as an Doomsayer it isn't true every time that Manglik marrying a non Manglik would lead to separation or death of the other consort. This lead me to delve further and I've come up with cases (obviously from the Book) when this yoga gets cancelled yea when Mars sits in the antecedent mentioned houses. I'll come to that discussion in the ensuing part of the essay but for now I would like to argue why the above mentioned houses are considered ill for marriage. 

1st house or Lagna is 7th from 7th. 7th is the Lagna for the nuptial mate and 1st house is makara house or end of life for the nuptial mate. 

 
 2nd house is the 8th house from 7th. As a result it shows risk and difficulties to the mate. 
4th house denotes domestic terrains. According to scholars evil world involving 4th or 8th from a bhava spoils the bhava. Hence Mars in 4th is evil with respect to health and things of the native. 
 
 7th house shows the legal yoke and health, time and characteristics of the spouse. So Mars in that house isn't conducive to the succeeding benefits. 

8th house shows difficulties of the native and finance and fortune of the mate. 8th house is also called Mangalya Sthana or duration of marital life so Mars there isn't good for marital life. 
 
12th house indicates the real pleasure which an integer have after marriage. Mars position presently's not conducive to happy marital life.

 Mars is in Scorpio (Vrischika) or Aries (Mesha) there's no dosha 
 Mars in Venus sign can not do injury if it's the 4th or 7th house.
